Identification of an immune modulation locus utilising a bovine mammary gland infection challenge model

Inflammation of the mammary gland following bacterial infection, commonly known as mastitis, affects all mammalian species. Although the aetiology and epidemiology of mastitis in the dairy cow are well described, the genetic factors mediating resistance to mammary gland infection are not well known, due in part to the difficulty in obtaining robust phenotypic information from sufficiently large numbers of individuals. To address this problem, an experimental mammary gland infection experiment was undertaken, using a Friesian-Jersey cross breed F2 herd. A total of 604 animals received an intramammary infusion of Streptococcus uberis in one gland, and the clinical response over 13 milkings was used for linkage mapping and genome-wide association analysis. A quantitative trait locus (QTL) was detected on bovine chromosome 11 for clinical mastitis status using micro-satellite and Affymetrix 10 K SNP markers, and then exome and genome sequence data used from the six F1 sires of the experimental animals to examine this region in more detail. A total of 485 sequence variants were typed in the QTL interval, and association mapping using these and an additional 37 986 genome-wide markers from the Illumina SNP50 bovine SNP panel revealed association with markers encompassing the interleukin-1 gene cluster locus. This study highlights a region on bovine chromosome 11, consistent with earlier studies, as conferring resistance to experimentally induced mammary gland infection, and newly prioritises the IL1 gene cluster for further analysis in genetic resistance to mastitis.

Macromolecular X-ray crystallography underpins the vigorous field of structural molecular biology having yielded many protein, nucleic acid and virus structures in fine detail. The understanding of the recognition by these macromolecules, as receptors, of their cognate ligands involves the detailed study of the structural chemistry of their molecular interactions. Also these structural details underpin the rational design of novel inhibitors in modern drug discovery in the pharmaceutical industry. Moreover, from such structures the functional details can be inferred, such as the biological chemistry of enzyme reactivity. There is then a vast number and range of types of biological macromolecules that potentially could be studied. The completion of the protein primary sequencing of the yeast genome, and the human genome sequencing project comprising some 105 proteins that is underway, raises expectations for equivalent three dimensional structural databases.

Since the emergence of HIV and its counterpart,acquired immunodeficiency syndrome (AIDS), rare opportunistic organisms have become increasingly prevalent. This increased prevalence has provided an opportunity for ultrastructural evaluation of these infectious organisms. Herein is presented a case of intestinal Mycobacterium avium-intracellulare (MAI) in a middle-aged male with uncertain HIV status at diagnosis.
A 56 year-old black male presented to University of South Alabama Medical Center complaining of shortness of breath, anorexia, nausea and vomiting, six month weight loss, and bloody diarrhea. His evaluation was negative for stool ova and parasites and fecal white blood cells. Stool culture and hepatitis viral panel was negative. Iron deficiency anemia and multiple calcified granulomas in both lungs were identified. On his third hospital day, he developed an acute abdomen. At exploratory celiotomy, segmental resection of jejunum and a liver biopsy were performed. Post-operatively the patient's cardiopulmonary status steadily declined and he died three days later.

National External Quality Assessment Schemes (NEQAS) in Microbiology have been established and operative in the United Kingdom for over 20 years. The essential components of such schemes with an account of the supporting expert advisory structures are described. Participation is voluntary and the primary aim is educational and designed to elevate standards of diagnostic practice generally through comparisons of performance within peer groups. The effect of such schemes on microbiological diagnostic performance is discussed.
The need for harmonisation within the European Community is addressed, together with an account of preliminary work relating to formulation of Directives in the field of medical devices.
The process of accreditation of diagnostic pathology laboratories through CPA (UK) Ltd. is briefly described.
Application of Analogue Amorphous Silicon Memory Devices to Resistive Synapses for Neural Networks

The amorphous silicon memory device shows promise as an analogue weight element in neural networks. The device resistance can be programmed to within 5% of any specific value between lkΩ and lMΩ using 10ns to 1μ voltage pulses in the range 1–5V. In this paper we describe the physical structure of the element and its electrical characteristics. Finally, a simple example is discussed of a small neural network implementing the EXOR function using amorphous silicon memory elements as a resistive array of weights and external op-amps as the current summing nodes.

Both Cr/p+/V and Cr/p+/Cr devices exhibit fast electrically programmable memory switching with a programmable voltage range, ΔVS, which depends only on the choice of the top metal. Despite a difference in ΔVS, both types of device have very similar electrical properties, but show one major difference in room temperature I-V characteristics, where under certain conditions, discontinuities are observed.

When a defence of insanity is put forward in answer to a criminal charge amnesia for the crime is not infrequently affirmed, in fact East (2) states that it is more often alleged than any other mental anomaly. The defence of amnesia is at times raised when there are no grounds for supposing that the prisoner is suffering from loss of memory, but there are undoubted cases where the crime, generally homicidal in character, has been committed during a disorder of consciousness or the memory of it subsequently has been lost. It is therefore of the greatest importance from a medico-legal point of view to distinguish between real and simulated amnesia. While amnesia may be simulated in cases where there is no mental disorder present, there are also instances where loss of memory is feigned by individuals in whom there are other reasons for regarding them as mentally abnormal. In this connection it must be realized that memory also is sometimes malingered; a prisoner who by reason of insanity accompanied by amnesia has escaped the responsibility for his crime may presently arrive at the conclusion that a persistence of his loss of memory will tend to keep him confined in an asylum, and therefore he asserts a remembrance of his crime, the story of which he is likely to have become familiar with at his trial or elsewhere. Not all such false memories, however, are definitely malingered. A patient may have heard the story of his crime so often that in the end he becomes convinced that he actually remembers it.
